Ideas to rid my shed of a stubborn squirrel?

You need to kill the squirrel. In a humane manner. This means no antifreeze. They make a block of poison bait called Tomcat. Squirrels seem to like it. It is much faster and less traumatic the antifreeze. Some squirrels do not like moth balls.

Go get a 12 pack of beer. Get a lawn or garden chair. Get a 410 shotgun or a pellet rifle with dialed in scope. When he shows himself, boom and go dig a grave for him. I have had a squirrel try and get in my attic. 4 gone so far and I think that they have given up for now. Get rid of them quickly is my opinion.

Why not just empty the shed and spray him out with a hose and then keep the shed closed from now on?
I must have dumb squirrels because a large rat trap with peanut butter worked every time.

mine were probably more interested in the tomatos I had growing than the peanut butter. I still cannot believe that I never caught one.

What kind of rat trap? Due to ongoing problems with squirrels getting in the attic, I recently bought some old-style wooden based Victor rat traps. Modified them by increasing tension on the spring with a spacer & drilling holes in the wooden base(a few vids on youtube about this). Then used 2 drywall screws apiece to mount 4 traps on several trees around the house, baited them with peanut butter with a little dry oatmeal mixed in. Some were sprung, one not, but on the sprung ones never saw any trace of hair. I did see evidence of bait eaten on 2 traps. Somewhere we have an old Havahart trap large enough for a racoon, may try that one soon, wire it open for a few days so they get used to free goodies. I took the rat traps down today, but can put 'em back up easy enough, cordless drill & drywall screws make it fast & easy.
BTW- while the Victor rat traps should be big enough for the gray/cat squirrels, I have serious doubts about them being big enough for the fox/red squirrels. We have both, plus occasionally some black squirrels show up.
Any trapping suggestions appreciated. I *do* have suitable pellet guns &/or .22 rifle or .410 shotgun, but it's very difficult to catch 'em when I can only watch 2 sides of the house at once. They just go to the other side of the house.

Modern coolant has a bittering agent added to it, so that won't work.
Rat poison and nuts/peanut butter should do the trick, or just a live trap with the same inside.

Not all coolant has Bitrex added, even the new stuff, so it might be worth shopping around..terribly cruel to the animal, though.

Just don't use CB Longs. I used those as a kid with the 29 grain solid nose, they are WAY too powerful around houses, also it is an actual firearm.
Pellet rifles have come a LONG way for pest control in North America in the last 15 years.
There is a world of Magnum Air Sporters beyond $$$RWS$$$ now.
But, one day when I am rich, I will own EVERY RWS air rifle in ALL CALIBERS.
But that is just the big kid in my talking... Maybe BITOG needs a RWS section??

Didn't read all of the answers but the supposed solution is to hook a strobe light to a motion sensor. Supposedly squirrels hate strobe lights. I think I heard this from car talk.
An owl would catch and eat the squirrel quickly.

OP, I too and not one for inhumane ways of getting rid of animals, but they are being pests. Damaging property. Not much you can do in a situation like this.
At least here, if you aren't in city limits and if a squirrel is being a pest, you can shoot it by law with no permits etc. Nothing wrong with that whatsoever IMO.

That was my first thought too. Whatever it is, is mighty quick. The modified traps shut harder & faster than when stock, & I found no trace of hair, feathers, blood or anything else. I may wind up sitting up on the off side of the roof with the Sheridan all pumped up & ready.

Do you have electricity in the shed? We had a problem with squirrels in our chicken coop, they were getting in and eating all the chicken food. We bought a sonic rodent repeller from Home Depot and have not had a squirrel problem since. Doesn't bother the chickens. It should get them out of your shed and keep them out.

That is the trap you need. Put a giant smear of peanut butter on the trigger. You'll have him in a day.

Look up a Victor Conibear 110 trap. These will definitely solve the problem much better than a rat trap. They can be a bit harder to find but worth looking for.
