Some thoughts:

What does the plug in #6 look like? Is it oil fouled?

Or is there an indication of a coolant leak from a bad head gasket?

You also might want to change the fuel filter and flush the fuel lines in case you have some fine sediment that is getting through and clogging this injector. I had that happen on a '97 Chevy pickup.

Check for vacuum leaks. Sometimes vacuum systems don't use vacuum equally from all cylinders. Vacuum hoses are Ford engines are usually quite weak. Also replace the PCV valve if it has over 20,000 miles on it.

You should do a compression check and see if 6 (And other) cylinders have low compression.
Many Ford V6 engines have head gasket problems.

Maybe a bad connection at the coil or the injector, try moving the connectors and wiring with the engine running.
If that doesn't help put a noid light on the injector connector and see if its getting juice when the engine is running, if it is look closer at the coil connector and associated wiring.
