I was mostly just wondering if it's possible to have a tiny hole in the rubber as a manufacturing defect. Does that ever happen?

What usually happens is that the tire is damaged in shipping.

The stuff that happens in the manufacturing plant generally gets caught in the plant because the finished tire is inflated several times before it goes into the warehouse.

Not to say the system is perfect, but leaks don't develop over time due to a manufacturing condition - they are either there from the "git-go" or not.
