Most of it comes what is called SmartPost. FedEx is responsible for getting it to a local Post Office (or the local USPS sorting facility), then your local Post Office delivers it whenever they feel like it... within a few days.

I also noticed an option when ordering test kits from Polaris that the UPS postage was $12 to a business address and $17 to my house. I thought business vs residential only mattered for freight!

I was trying to order a case for it. I'm not sure why, but the prices for EVERYTHING are showing up $20 higher than on the web site?
$12.99 for a case looks good - I added it to a case and suddenly it's $32.99
Same with screen protectors. $2.99 but when I added them to the cart, they were $22.99 ?
Are they jacking their prices up $20 on everything for those who are not amazon prime members?

No, but you need to look at who the seller is.
Also, this increase should not come as a surprise. The $25 threshold has been there for a decade, and Amazon warned us earlier this year that it would go up for the holidays.
What's worse is that they are building a new distribution center in Baltimore, which means that I will soon have to pay sales tax as well. Stuff now is often shipped from PA (near Harrisburg) will now only come across town. So instead of coming to my house practically overnight, it will come practically overnight instead. My point being that the stuff won't get to my house any quicker, and this only increases my costs.

I have prime so I didn't notice, most things I buy are shipped for free second day.
I noticed that Tuesday night. I've also had the privelege of paying sales tax for over a year now.
