Is there a general rule that on an auto transmission the rear-most fluid line is the return line (from the radiator) and the front-most is the supply line (to the radiator)? I'm going to install a transmission cooler and am just trying to make sure. Thanks in advance.

Allow your engine and transmission to completly cool down. Have someone set in your car with their foot on the brake and have the parking brake set! They can start the car and put it in drive. While they are doing this, feel each of the hard lines from the transmission and see which one gets hot first. The other "cooler" line is the return.
Watch out, though, it can squirt 40 feet!
Don't take this as a hard-and-fast rule, but typically fluid should flow from the bottom of the cooler to the top, so as to eliminate air pockets. You can use that as a hint, but the definitive information should come from a service manual or by pulling a line.
You can disconnect the ignition coil wire, disconnect one cooler hose, crank the engine, and see the direction of flow of the fluid. The transmission does not need to be in drive.
