In a dry NOS system, like NOSzles that fit under the fuel injector, would the NOS be producing a positive pressure environment in the intake manifold runners?

No, I don't think so. The intake is still under vacuum unless there is some sort of forced induction. (I might be wrong.)

No, there's not enough volume of gas coming from the nitrous system to overcome the intake vacuum. It takes a great deal of volume to overcome that and nitrous jets are pretty tiny, relatively speaking.
