How does Auto-RX rejuvenate seals?

My friend John has a 1997 Ford F150 4x4 and transmission fluid is leaking into the transfer case.

I thought this might be a problem that Auto-RX could be the solution for, but he asked me today how Auto-RX works and I really couldn't give him any answer other than that it makes the seals more pliable so that they can conform to their mating surfaces better. I did tell him that it fixed a rear main seal leak on my 1996 Contour.

I realize that this probably has something to do with the effects of esters on seals, but what exactly do esters do to seals?

Also does Auto-RX only work on rotational seals like rear main seals or does it work on seals like valve cover seals and oil pan seals? (Maybe not as well on those?)

Auto-Rx will not have an effect on valve cover and oil pan gaskets as a normal. Especially if they are cork or silicone.

On metal parts the Auto-Rx cleans the metal and also cleans the seals. This cleaning allows the Auto-Rx to better conform to the metal and help prevent leaks. It does not swell the seal, it just cleans it and makes it more pliable. Probably not the best explanation, but basic.

So basically if there's a bit of dirt buildup under the seal which is preventing it from sealing, Auto-RX will remove this dirt and allow the seal to work correctly?

Yes, it will remove the sludge and varnish to let the seal conform properly.
