Will it make a significant difference over the life of the oil, probably no.

Is there are real difference, definitely yes.

A full synthetic 0W-20 is a great oil, just see Tig1's post above, but if we are here to split hairs (and most are) and it's above freezing and both oils are formulated the same, then the 5W20 does have better properties and they are measurable.

The oil formulator Joe believes there is a relation between sticking piston rings and oil volatility, with lower oil volatility being better.

You could buy 2,000 Prii, and load 1000 of them in 0E-20 format, the other 1,000 in 5W-20 format, run them for 200,000 miles;
Disassemble all of the engine parts and measure all of the wear.

My bet would be there would be no statistically relevant difference between the 2 fleets.
