I have been searching for 30 minutes and cannot find the engine coolant block drain for my '19 ES350. I understand there is only 1 block drain. I have seen pics of it being on the driver's side. I have read posts where it is on the passenger side. I you could just help to narrow down my search to either driver or passenger side, I would be very grateful. 2GR-FKS engine

I have no idea. I just found this one. You will have to obviously get under and search for it. Even on my 5.7 that has two block drains on each side, it is far easier to access; when you look for them, it is very small and hard to see at first glance.
They're tucked under the exhaust manifolds.; IIRC.
The block drain has to be near the bottom of the block or it's not a drain. It will not be much more than 2" or so inches above where the oil pan is bolted to the block.
Drain plug likely to be brass. Get some PB Blaster in there a few minutes before you start. No idea if it's hex or square. If square then an 8 pt socket would be best.

A guy on Sienna Chat described the coolant drain on the 2GR-FKS as being near the rear of the engine (next to the transmission) on the firewall side.

All Toyotas drain plugs that I own have a 10 mm nut. I found one on the back side to the left of the exhaust manifold. I am assuming
there is only the one block drain. The Lexus service mgr texted me there were two on the block. One on front and one on the rear.
I haven't heard back from him as of this moment.

Unless you are doing a cleaning with a heavy duty cleaner and really need to flush very well or are changing coolant chemistry then I don't think you really need to drain via block drain plug.
I went from Ford orange to Ford yellow in my PSD. While compatible you don't get the full benefit of the yellow coolant unless you get all the orange out. So I had the Indy mechanic drain via drain plugs. And to make it even more of a PIA my PSD has two complete cooling systems.

The voices, man... if they say you need to do it, you need to do it. 
You’re asking for more trouble than not by opening that block drain. Just drain radiator, add pre-dilute coolant, get engine to operating temp, drain one more time, then refill with pre-dilute.
The block drain in the 2gr-fks is #3 in this diagram from Mitchell service data. Service data does recommend draining the block, but do not describe its location. From the image, looks like driver's side, back of engine as someone previously mentioned.

Sorta hard to tell from the pic. I finally found it. Passenger side on rear bank. Easy to get to. Never found the other block drain. I’m assuming there isn’t one. Even though service mgr at Lexus said there were 2 block drains on this engine???
