Best way to sample manual fluid

Is the best way to sample a manual trans fluid to run the car, get it up to operating temp, and just take a sample from the middle of the drain flow like you would engine oil? Just wondering since there is no oil pump, if things tend to settle on the bottom of the transmission that would fill up your sample?
 
It'll take a while to settle. So, just like you said, get it warmed up and grab a sample after a pint-quart drains out.
 
I couldnt in my saab. The drain plug is next to a suspension crossmember such that it is impossible to open it up. I sucked out my MTF via the fill hole.

To get a sample I used an oil analysis vacuum sample kit, which worked great. Car was run and warmed as much was possible, though surely settlement occurred by the time the car was lifted, wheel off, plug out and sample taken.
